Vaccine Storage and Handling Requirements for Pharmacy Technicians

Master CDC vaccine storage guidelines, temperature monitoring, and cold chain management essential for the PTCB exam and daily practice.

Why Proper Vaccine Storage Matters

Improper vaccine storage can reduce potency or render vaccines completely ineffective. Pharmacy technicians play a critical role in maintaining the cold chain from delivery to administration. This topic is tested on the PTCB exam and is vital for daily practice.

Temperature Requirements

Refrigerated Vaccines

Most vaccines require storage between 2 and 8 degrees Celsius (36 to 46 degrees Fahrenheit). Examples include influenza, hepatitis B, and DTaP vaccines. Never store vaccines in the door of a refrigerator where temperatures fluctuate.

Frozen Vaccines

Some vaccines, such as varicella (Varivax) and MMRV (ProQuad), must be stored at minus 50 to minus 15 degrees Celsius. Once thawed, these vaccines cannot be refrozen and must be used within specific timeframes.

Storage Best Practices

  • Use purpose-built pharmaceutical-grade refrigerators and freezers
  • Place a calibrated digital data logger (DDL) in each storage unit
  • Record temperatures at least twice daily, at the start and end of the workday
  • Keep vaccines in their original packaging to protect from light
  • Maintain at least 2 to 3 inches of space between vaccine containers for air circulation

Handling Temperature Excursions

When a temperature excursion occurs, do not discard vaccines immediately. Mark them as "do not use," store them at the correct temperature, and contact the vaccine manufacturer or your state immunization program for guidance on viability.

Emergency Preparedness

Pharmacies should have an emergency vaccine storage plan that includes backup storage locations, portable coolers with conditioned water bottles, and staff notification procedures for power outages.
